Abstract:Single factor and orthogonal experiment methods were used to explore the tissue culture technique of Hemerocallis citrine ‘March Flower’. The results showed that the best effect was when the young leaves were treated with 0.1 % mercuric chloride for 12 min with no pollution and death and the activation rate was 62.5 % after 14 days. MS + 2 mg/L 2,4-D + 0.1 mg/L 6-BA was the optimal induction medium with the 86.67 % recovery rate, MS + 2 mg/L 6-BA + 0.5 mg/L IBA was the best differentiation medium with the 80 % differentiation rate, and the average number of adventitious buds per block was 1.72. 1/2 MS + 0.5 mg/L NAA was the best rooting medium, the rooting rate was 93.33 %, the average number of rooting per shoot was 5.83. The survival rate of transplanted test tube seedling was 95 %. Using the tissue culture technique, the young leaf explants obtained the "March flower" daylily regeneration seed through the callus pathway.
