      Abstract:We characterize the hypercyclicity of weighted composition operators on the Little Bloch Space and the Besov space.We obtain that there are no hypercyclic composition operators on the Little Bloch Space and the Besov space when holomorphic self-map is an automorphism.And when holomorphic self-map is a non-automorphism,we discuss the hypercyclicity of weighted composition operators on the Little Bloch Space and the Besov space under certain conditions.

      Abstract:In this paper, we discuss the disjoint hypercyclicity of linear composition on the weighted Banach spaces.Moreover,according to the difference of the analytic maps,we obtain a sufficient condition for the disjoint hypercyclicity and disjoint supercyclicity of composition operators on $H^\infty_{\alpha,0}$. Moreover,we also obtain a partial characterization for the hypercyclicity of weighted composition operators on $H^\infty_{\alpha,0}$.

    • Cubic Trigonometric B-spline Collocation Approach for Black-Scholes Method

      Abstract:A cubic trigonometric B-spline collocation approach is developed for the numerical solution of Black-Scholes equation governing European option pricing. The Black-Scholes equation is fully-discretized using the cubic trigonometric B-spline collocation for spatial discretization and the forward finite difference for the time discretization. A hybrid difference scheme is obtained by means of parameter θ. According to Von Neumann (Fourier) method, it is shown that the presented scheme is unconditionally stable for 1/2≤θ≤1. A numerical experiment is performed to illustrate the validity and accuracy of the proposed method. Moreover, the numerical results are given to show that it is superior to Crank-Nicolson finite difference method and cubic B-spline collocation approach.

      Abstract:In this paper, the authors propose a new stabilized finite element formulation for the incompressible time-dependent Navier-Stokes equations with high Reynolds number. This formulation combines subgrid eddy viscosity methods with H(div) finite element approximation, for example RT and BDM finite element. This method not only satisfies the conservation condition but also controls spurious oscillations in the velocities due to the convection dominated. We derive the stability and error estimates for finite element semidiscrete scheme which combines subgrid scale eddy viscosity method with \textbf{H}(div) elements. In addition, the constants in these error estimates do not depend on the Reynolds number but on a reduced Reynolds number.

      Abstract:We study the relation between certain rational surfaces and orthogonal Lie algebras (that is, $D_n$-Lie algebras). We find that a fundamental irreducible representation (whose highest weight is denoted by $\lambda_{n-2}$) is determined by finitely many rational curves on these surfaces satisfying two systems of Diophantine equations, and the solutions of each system of these equations form a Weyl group orbit.

      Abstract:In order to solve the problems of trival wiring and limited collection scope existing in the process of traditional crop image data acquisition, a mobile remote image acquisition system was designed. With cortex-A8 architecture processor S5PV210 as the core of the system,the UVC camera was selected for the image acquisition device, and the telnet or gravity of the phone's screen induction reaction mode was used to pass parameters to the background through CGI (common gateway interface) , then the mobile device control and on-board PTZ (Pan/Tilt/Zoom) equipment operation were realized. In control interface the images were not only acquired over long distance on grapery, but also the monitoring areas were adjusted by remote controlling the mobile device through the developed program. The performance experiments of the mobile acquisition system had been implemented in the vineyard of Gansu Agricultural University. The results showed that the system operation credibly, laid lines concisely, and image transmission stably. While in telnet terminal control mode,the system has shorter time response, better real-time, longer transmission distance, and lower frame loss rate etc. This kind of research and development provides a new intelligent prototype equipment for vineyard information monitoring, and which is of great significance to design and develop agricultural IOT(Internet Of Things) and its application.

      Abstract:In order to improve CV model’s performance on image with texture, we analyze the statistical properties of image texture components, and propose an improved segmentation algorithm. Specifically, this study has designed a new edge-preserving function based on the TV image smoothing. Also, the algorithm applies the CV model to segment the smooth component of an image. On the other hand, considering the fact that different smooth components have different segmentation areas, a new Smooth convergence function based on the region confidence is designed to overcome the disappearing of segmentation curve. Experimental results demonstrate that the proposed algorithm outperforms the traditional CV model in term of segmentation effects on natural images, but is insensitive to the inconsistent area.

    • Successive Block Diagonalization Precoding for Multi-User MIMO System

      Abstract:Block Diagonalization (BD) is a common linear precoding technique for the downlink of multi-user MIMO(MU-MIMO) systems. A drawback of conventional BD is that the effective channel gain for each data stream can be severely unbalanced when each user has multiple data streams. In this paper, a new precoding scheme called successive BD (SBD) is proposed based on BD precoding by introducing a new construction scheme of interference matrices considering inter-antenna interference. The precoding matrix responding to each of the receive antennas is successively calculated. SBD precoding reduces the difference in effective channel gain between each user’s multiple data streams caused by the original BD precoding and improves the average bit error performance of the user. More spatial diversity gain can be obtained compared with BD precoding when precoding and combing are employed and spatial diversity transmission strategy is adopted. Simulations show that when the bit error rate is 10^(-3), the proposed method has about 2 dB gain over the conventional BD precoding in {2,2}*4 MU-MIMO systems.

      Abstract:User attributes play an important role in personalized service. The prediction of the user's property based on mobile phone data has gradually become a new direction. In this paper, we use two independent attributes: average daily usage time and number of application categories. The basic attribute and the concept of the auxiliary attribute are proposed. In this paper, firstly, the auxiliary attributes of all unlabeled samples are discretized by non-supervised method. And then calculate the Hellinger Distance of auxiliary property categories, which is the characteristic weight of the basic attribute. Input the basic attributes and the characteristic weight to the base classifier of the ensemble classifier training model, introducing random forest with out of sample accuracy as the base classifier weights, finally we get the final classification results. The experimental results show that the ensemble classifiers framework can improve the effect of user attribute prediction.

    • Joint Angle, Range and Velocity Estimation for bi-static FDA-MIMO Radar

      Abstract:It is known that there is a contradiction between range estimation and velocity estimation due to the influence of pulse repetition frequency (PRF) in traditional phased-array (PA) radar. In this paper, a joint unambiguous angle, range and velocity estimation method is proposed for bi-static multiple-input-multiple-output (MIMO) radar with frequency diverse array (FDA). Owing to the FDA, there is a little frequency increment between the arrays and the transmit steering matrix carries angle, range even velocity information. Consider the range-angle coupling in transmit steering matrix, a subarray-based transmitted waveform and decoupling approach are designed. Utilizing the ESPRIT algorithm and ambiguity resolve methods based on FDA, the direction-of-departure (DOD), direction-of-arrival (DOA), range and velocity parameters are estimated unambiguously. Additionally, Cramér-Rao bounds (CRB) for these parameters are derived and the performance of this radar is analyzed. Simulation results indicates the validity of the proposed method.

    • Design and Simulation of the Reconfigurable Surface-wave Waveguide Based on Petal Slotted Tunable High Impedance Surface Cells in S Band

      Abstract:The surface-wave waveguide, based on the high impedance surface, is widely used to transform the surface-wave energy or to guide surface-waves along a constrained path. However, there still remain two issues for almost surface-wave waveguides. First, the guided-wave path is fixed, which will prevent changing the guided-wave path in real-time situation. Second, the working frequency band is usually high, which is in X band or higher frequency range. The two issues have limited the large-scale applications of the surface-wave waveguide, especially for the first one, which means that one has to design a waveguide for every guided-wave path, leading to the high cost. In this paper, a high-impedance square cell with petal slots attached to four varactors (SMV1408) is designed, its impedance value increases from j390Ω to j1710Ω in S band by decreasing the reverse bias voltage of varactor from 20V to 0.2V. Based on this tunable cell, we propose the concept of the reconfigurable surface-wave waveguide and its feasibility has been proved by HFSS. In the simulation, the proposed waveguide can provide multi-guided paths and the paths can be adjusted in real time; meanwhile, to prevent the energy leak of surface-waves, the guide-wave path is optimized by our previous achievements, and the effectiveness of our suggestions is verified well by the simulation results.

    • Design of a miniaturized rectifying circuit with a via-hole interconnected multilayer structure

      Abstract:This article proposes a novel concept for constructing a microwave (MW) rectifying circuit by using a multilayer structure, aiming at reducing physical dimensions. Specifically, conductor layers in the multilayer structure are interconnected with an embedded via-hole. Then, an S-band rectifying circuit based on HSMS-282C Schottky diode is fabricated with the proposed multilayer structure for demonstration. The maximum MW-DC conversion efficiency of 73% has been measured. In comparison with the conventional microstrip rectifying circuit, the proposed multilayer circuit shows a great reduction of physical dimensions and a very similar efficiency, which validates that the proposed circuit can reduce physical dimensions significantly without sacrificing the MW-DC conversion efficiency.

      Abstract:In the detection of maneuvering weak targets, the statistical characteristics are unknown and the echo signal is difficult to model for the clutter and interference in the echo signal, so the posterior probability density function can’t be obtained and the performance of traditional particle filter algorithm decreases. Aiming at this problem, target detection before track algorithm with MIMO radar based on cost reference particle filter. The algorithm does not require of the statistical characteristics of the background noise. The difference between the true value and the estimated value of the target state is used to calculate the cost and weight of each particle, the posterior probability density doesn’t require the model of noise. Simulation results show that the proposed algorithm is superior to the traditional particle filter algorithm when the noise statistics are unknown.

      Abstract:In this work, the experimental techniques of supersonic molecular beam, femtosecond laser and time-of-flight (TOF) mass spectrometry were applied to investigate the photoionizations of nitrogen and oxygen molecules under intense femtosecond laser field. By way of analyzing the TOF mass spectra and yields of fragment ions, we got the dependence of ions on the laser intensity, laser ellipticity and the laser polarizations. In combination with intense field theory, we analyzed the photoionization mechanism in detail. 

    • Elastic and thermodynamic properties of LaB6 under pressure: a first-principles study

      Abstract:The elastic and thermodynamic properties of CsCl-type structure LaB6 under high pressure are investigated by first-principles calculations based on plane-wave pseudopotential density functional theory method within the generalized gradient approximation (GGA). The calculated lattice parameters of LaB6 under zero pressure and zero temperature are in good agreement with the existing experimental data and other theoretical data. The pressure dependences of the elastic constants, bulk modulus B (GPa), shear modulus G, Young’s modulus E, elastic Debye temperature ΘE, Poisson ratio σ, compressional wave velocity VL and shear wave velocity VS are also presented. An analysis for the calculated elastic constants has been made to reveal the mechanical stability of LaB6 up to 14 GPa. The thermodynamic properties of the CsCl-type structure LaB6 are predicted using the quasi-harmonic Debye model. The variation of thermal expansion coefficient α, the variation of the heat capacity Cv are obtained systematically in the ranges of 0-14 GPa and 0-1500 K. At last, the pressure dependences of the density of states are also investigated.

    • Study on Structural Stabilities and Electronic Properties of Medium Size ZnSe Clusters

      Abstract:Structural stabilities and electronic properties of medium sized (ZnSe)n(n = 24, 28, 36, and 48)clusters are investigated in this paper. The starting configurations are generated by handmade construction or cutting from bulk material. Some low-energy structures of other II-IV clusters are also considered. The structure optimizations and energy calculations were performed using DMoL package. The results show that the energies of tubular and cage structures are the lowest when n=24, 28, and 36, and the energy of wurtzite (WZ) bulk material is the lowest when n=48. The tubular and cage structures have relatively large HOMO-LUMO gaps, whereas for the onion-like structures and the WZ bulk materials, their energy gaps are smaller than those of the cage and tube structures.

      Abstract:In this work, structural stability, band structure, density of states and charge density difference of iron, cobalt and nickel doped single-layer phosphorene are studied by first principles method. The research of the thesis can be mainly divided into following results, the adsorption stability of adsorption in top at phosphorus position of cobalt is stronger than that of iron and nickel in the adsorption system; as well as the stability of Ni doped system is the strongest in the doping systems. The adsorption of iron, cobalt and nickel on the surface of phosphorus can be better regulate the energy band structure, so we can get controllable performance ofsemiconductor materials. The band gap values were 0.52 eV, 0.56 eV and 0.4 eV as iron, cobalt and nickel doped singlr-layer phosphorene, respectively.Nearest neighbor phosphorus atoms appeared electronic aggregation phenomenon at around iron, cobalt and nickel doped; because of iron, cobalt and nickel have two electrons in 4S orbit and phosphorus atoms are easier to get electrons

      Abstract:Studies aimed at understanding the nonlinear optical properties of GaAs/AlGaAs semiparabolic quantum well under applied electric have focused on optical rectification and second harmonic generation. These studies have taken two complimentary approaches: (1) the compact-density-matrix approach and iterative method have been used to obtain the expressions of optical rectification and second harmonic generation. (2) the finite difference techniques has been used to obtain the energy eigenvalues and their corresponding oeigenfunctions of the semiparabolic quantum well under applied electric field. The energy eigenvalues, the shape of the confined potential, optical rectification and second harmonic generation are modulated by the confined potential frequencies, and electric field. So the results of a number of numerical experiments indicate that the nonlinear optical rectification and second harmonic generation strongly depends on the confined potential frequencies, and applied electric field. This gives a new degree of freedom in various device applications based on the intersubband transitions of electrons.

      Abstract:The influence of PVA on the pyrolysis was studied from macro and micro angles. In this paper, briquette pyrolysis was operated with thermal analyzer. The experimental data showed that with the increase of PVA content, the R value, the reaction rate and the low calorific value increased. The FTIR spectrum, BET, industrial analysis and quantum chemical analysis were described that the addition of PVA was advantage to briquette pyrolysis. The number of micropores was increased and the heat and mass transferation was improved among the fuel layer with the addition of PVA.

    • Dynamic adsorption and elution performances of H2TiO3-lithium adsorbent loaded on ceramic foams

      Abstract:H2TiO3-lithium adsorbent was loaded on cordierite-based ceramic foams by the sol-gel process. The adsorption and elution of Li+ by the obtained samples were studied with the ion exchange columns via the dynamic process. The effects of liquid flow speed, Li+ concentration and single column/multi columns on the adsorption capacity and enrichment factor of Li+ were studied. The results indicate that it needs a long time for the lithium adsorbent loaded on ceramic foams to reach the leakage, and the adsorbent has a high operational adsorption capacity. In a certain range, flow rate of the eluent does not affect enrichment factor of Li+ apparently. The adsorbent does not have the ability to enrich Li+ with a high concentration. However, it is capable for the adsorbent to enrich a low level of Li+. The enrichment factor of Li+ can at least reach 26.55 times by adding hydrochloric acid to adjust pH of the effluent liquid to equal to that of the original eluent and using the obtained liquid as the eluent repeatedly.

      Abstract:Abstract: We calculate the equilibrium geometry structure and the anharmonic force field of OCN4 by using B3LYP, B3p86, B3pw91 and MP2 methods combining  with cc-pVnZ (n=D,T) basis sets. From the calculated results, we can see that the calculated values of DFT methods are very close to each other and excellent agreement well with experimental values. Comparing with the changes caused by methods, the basis sets playing the predominant role. Overall, the DFT methods combining cc-pVTZ basis set provide more reliable results of spectroscopic constants and anharmonic force field of OCN4.

      Abstract:In order to obtain high-performance batteries with titanium dioxide films by RF magnetron sputtering, doped(V2O5) TiO2 films were prepared. The main factors affecting the time,the pressure ratio of argon and oxygen, and annealing temperature. The effects such as mass of doping (V2O5) and annealing temperature on film properties.722 spectrophotometer using the optical properties of the study by the degradation of methylene blue and drop contact angle method on the catalytic properties of thin films and hydrophilic properties were studied. The results showed that Doped (V2O5) red shift the absorption peak of the film and the band gap narrowing, of which 0.5% V2O5 doped TiO2 thin film of the red shifts of the largest band gap becomes 2.88eV; 400℃ annealed films with good photocatalytic properties and ultra-hydrophilic.The prepared thin films of titanium dioxide as DSSC cells of sputtering time, the doping effect on battery performance. it shows that Doped with 0.5% V2O5 thin film battery open circuit voltage and short circuit current is higher than the undoped cell, in particular, short-circuit current increases from 24.82A to 88.15A.

      Abstract:The model of biomagnetite magnetoreceptions include the model of single-domain magnetite magnetoreception and the model of superparamagnetic magnetite magnetoreception. In honeybees, there are many typical superparamagnetic magnetites, so in this paper, the magnetic model and working mechanism of the superparamagnetic magnetite magnetoreception are discussed qualitatively from the point of view of theory of magnetism taking the example of honeybees. In the outer magnetic field, each superparamagnetic magnetite can be act on with the size extend or contract along different directions. Superparamagnetic magnetites transmit the force and change of size to the outer biomembrane and eukaryotic cytoskeleton which embedding in the nervous system, so a nerve signal can be produced. The signals can be magnified because many superparamagnetic magnetites exist.

    • The expression of long non-coding RNA LINC00941 in colorectal cancer and effect on cell proliferation

      Abstract:To explore the function of a cell differentiation-associated long non-coding RNA (lncRNA) LINC00941 in cancer progression, we detected the expression of LINC00941 in 6 types of human cancer cell lines and HEK-293 cell line by real-time quantitative PCR. LINC00941 is highly expressed in colorectal carcinoma cell lines HCT116 and HCT116 p53-/-, lung carcinoma cell lines A549 and NCI-H1299, and melanoma cell line Stilling, especially in colorectal carcinoma cell lines. By detecting the RNA levels of LINC00941 in the tumor and normal tissues of colorectal cancer patients, we found that the expression levels were significantly increased in tumor compared with paired normal tissues. By shRNA(short hairpin RNA) interfering, we found LINC00941-knockdown HCT116 cells had lower proliferation rate compared with negative control cells, suggesting the correlation of LINC00941 and colorectal cancer progression.

    • The Study on Different Extraction Methods and Antioxidant Activity of Volatile Oil from Saffron Bio-residues

      Abstract:The extraction of volatile oil was performed by water vapor distillation, ultrasonic and Soxhlet extraction from saffron bio-residue. 49 kinds of compounds were identified in total and the relative content in percentage were calculated with area normalization by GC-MS. Three methods can extract 43, 27 and 21 kinds of components respectively and share 6 components together. The main components are palmitic acid, linoleic acid, linolenic acid. In contrast, water vapor distillation can harvest showed the most variety of components such as linalol,eucarvone and cedrol etc.. What’s more, the relative content of safranal was 40.31%. Soxhlet extraction can show the highest extraction rate. In the experiment of antioxident activity, three extracts were tested by DPPH, reducing power and T-AOC for antioxidant measurement, respectively. The clearance rate of water vapor distillation extracts reached 44.17%, which was the highest in the three extraction methods when the steam distillation’s extract concentration at 280μg/ml. The extract by Soxhlet extraction showed the best result in reducing power and T-AOC tests.

    • cDNA cloning of heat shock protein 60 (SpHsp60) in Stegobium paniceum and its expression under temperature stress

      Abstract:The full-length cDNA sequence of a heat shock protein 60 (Hsp60) gene from Stegobium paniceum (a major insect pest in stored Chinese medicine material) was cloned by using RT-PCR technique, and termed as SpHsp60 (GenBank accession no. KX778623). Sequence analysis showed that SpHsp60 contained an open reading frame of 1719 bp encoding 572 amino acids, with a predicated molecular weight of 60.9 kDa and isoelectric point of 5.32. SpHsp60 had the highly conserved functional motifs of the Hsp60 gene family, and SpHsp60 shares 88% amino acid sequence identities with the Hsp60 proteins from Tribolium castaneum and Leptinotarsa decemlineata. The quantitative real-time PCR results indicated that the expression of SpHsp60 was significantly increased 2 hours after exposed to -5°C and 0°C, compared to the control, and this gene was also induced after exposed to 42°C. The results suggest that the SpHsp60 gene may correlate with extreme temperature-stress tolerance in S. paniceum.

    • An evolutionary modular view of functional diversity in Arabidopsis uridine diphosphate glucosyltransferase

      Abstract:Uridine diphosphate (UDP)-glycosyltransferases (UGTs), belonging to the family 1 glycosyltransferases (GTs), play important roles in transferring glycosyl moieties and glycosylating secondary metabolites in Arabidopsis thaliana. Reconstructed phylogenomic trees can provide novel insights in studying evolutionary relationship and functions of this multigene family. By combining comprehensive phylogenetic profiles and reconstructed phylogenomic trees, we systemically analyzed 112 Arabidopsis UGTs. In total, we detected five distinct evolutionary conserved modules, and most UGTs were divided into the same module indicating a rather conserved evolutionary routine of Arabidopsis UGTs. We next manually collected the substrates of Arabidopsis UGTs and mapped them to orthologous groups, revealing evolutionary relationships of UGTs in binding specific substrates. By scanning 271 different species, we also predicted several unreported proteins that were closely related to Arabidopsis UGTs.

    • The spatial and temporal variation of glutamate decarboxylase (Gad) property in Brassica rapa L. ssp rapa

      Abstract:In this study, crude enzyme extraction method, diversity property and light influence of glutamate decarboxylase(Gad) in Brassica rapa L. ssp rapa were analyzed. Ultra Performance Liquid Chromatography(UPLC) method was used to measure the production in Gad catalyzed progress-- Gamma - aminobutyric acid(Gaba). Adding 1% cellulase solution at homogenization step could improve the enzyme activity up to about 70% as much. Meanwhile, when pH were 4.5-6 with the temperature of 55℃,enzyme had reached its maximum activity ,on the other hand Ca2+ and coenzyme Plp didn't have any contribution. Among the different developing stages of Brassica rapa L. ssp rapa, extration of sucking seedling apical meristem could generate more Gaba. In general, with proper conditions, the sucking seedling could be the most efficient material and easily obtained, thus having considerable prospects of industrial use.

    • Study on the optimization of Ophiopogon japonicus Fermentation and the Moisture Retention

      Abstract:Abstract: Objective: To investigate the effects of different fermentation conditions on Ophiopogon japonicus fermentation liquid moisturizing function, to obtain the optimum fermentation process. Method: Single factor test was used to adjust fermentation parameters such as the preparation method of fermentation substrate, Ophiopogon japonicus concentration, inoculum, fermentation temperature, fermentation time and rotational speed with the moisture retention of the Ophiopogon japonicus fermentation liquid. Optimization of fermentation parameters of Ophiopogon japonicus based on response surface method, studied the moisture retention of the obtained fermentation liquid of Ophiopogon japonicus. Result: The optimum fermentation process of Ophiopogon japonicus liquid was obtained,and the fermentation liquid of Ophiopogon japonicus has the better moisture retention. Conclusion: Ophiopogon japonicus fermentation liquid has good moisturizing properties and can be used in cosmetic raw materials.

    • Effect of Rape Straw Ratio on Performance of Aerobic Composting Process and Characteristic of Final Product

      Abstract:Composting is an effective method to convert crop straw into organic fertilizer. To maximum utilize rape straw, the effect of rape straw ratio on performance of aerobic composting process and final product was investigated. The rape straw proportion in feedstocks in R1, R2 and R3 were 45, 65 and 85% (wet weight), respectively. The results showed that organic matter degradation in R1, R2 and R3 occured mainly in the early stage of composting process, and achieved 34.06, 35.57 and 39.78%, respectively at the end of composting process. In addtion, the degradation of organic matter conformed to the first-order kinetic model. Comparing the physicochemical properties of samples during the composting process, the end-product of R3 satisfied all the maturity indicators recommended by previous references. In the end-product of R3, NH4+ was not detected, and the total nitrogen content, NO3- concentration and germination index (GI) achieved 2.17%, 1,844.93mg•kg-1 (dry weight) and 102.66%, respectively. In conclusion, rape straw with higher addition amount enhanced the utilization efficiency of rape straw, and enhanced the quality of the end-product. Moreover, nitrogen loss reduced with increasing rape straw addition amount in the present experiment scope.

    • Inhibitory effects of Salvia miltiorrhiza bge . f . alba water extract on apoptosis of islet B cells in rats

      Abstract:Salvia miltiorrhiza bge. f. alba is a kind of rare Chinese herbal medicine, which contains multiple kinds of trace elements and has high medicinal value. In this paper, MTT method was used to detect the inhibitory effect of Salvia miltiorrhiza bge. f. alba extract on the apoptosis of INS-1 cells, glycemic and insulin was tested, and HE staining and immunohistochemical staining was carried out. The experimental results show that Salvia miltiorrhiza bge. f. alba can promote the production of Bcl-2 proteins, weaken the ability of Bax proteins to promote apoptosis, thereby increase the release of insulin, ultimately achieve glycemic control purposes. The research of this paper has certain significance for the development of adjuvant drugs for the treatment of diabetes mellitus.

    • Elevation of immunity to PCV2 vaccine by recombinant Tibetan pig interleukin-12 gene in chitosan nanoparticles

      Abstract:To study the regulation effect on vaccination of porcine circovirus-2 (PCV2) by co-delivering interleukin-12 (IL-12) cDNA via chitosan nanoparticles, two subunits of IL-12, P40 and P35 of Tibetan pig, were cloned into mammalian expression plasmid VR1020. VRIL-12 was transfected into HEK293 cells and PCR test was conducted. The results showed that IL-12 gene could express successfully. The recombinant plasmid was packaged into chitosan nanoparticles using ionic crosslinking method. The nanoparticles (VRIL-12-CNP) were confirmed to be nanoparticle size with good dispersion and positive charge. VRIL-12-CNP was then used to inoculate the 21-day-old piglets together with PCV2 vaccine. Blood samples were taken at day 0, 7, 14 and 28 post inoculation for analysis. The results showed that post vaccination the T cells such as CD3+, CD4+ , CD8+, and anti-PCV2 specific antibody, all increased significantly in the VRIL-12-CNP co-delivering pigs (P<0.05), the expression levels of TLR2, TLR7, IL-12, IL-4, IL-6 and IL-15 in the co-delivering group were significantly higher than those in control group (P<0.05), and the expression of STAT1, 3 and Bcl-2 gene was also increased significantly in the co-delivering pigs (P<0.05). Moreover, the net weight gain of the pigs in co-delivery group was significantly increased in the 28 days post inoculation (P<0.05). These results indicated that co-delivery of VRIL-12-CNP elevated swine innate immune response and both humoral and cellular immune response to PCV2, and VRIL-12-CNP is a promising safe and effective adjuvant to PCV2 vaccination.
