The gastric inhibitory polypeptide/glucose-dependent insulinotropic polypeptide (GIP) shows the inhibitory effect on gastric acid secretion and incretin action. The aim of this study is to analyze the cDNA sequence of the pig GIP (pGIP) gene, and further examine the tissue expression of pGIP based on RT-PCR method. Result showed that pGIP cDNA is 532 bp in length and encodes a 144-amino acid GIP precursor which contains a signal peptide. RT-PCR assay showed that pGIP mRNA is highly expressed in kidney and gastrointestinal tract. Characterization of the pGIP gene will help to elucidate its physiological role.
